•Hierarchical constrained local model to locate anatomical landmarks proposed.•Statistical shape models using ICA to describe local shape variations.•A data-driven independent components selection method proposed.•Combination of geometric and texture features for genetic syndrome detection.•The method validated on a dataset including 130 images achieved 0.967 accuracy and 0.956 F1.
