•A multi-template framework is proposed to quantify perirhinal cortex (PRC) using MRI.•The framework explicitly models the 3 discrete anatomical variants of PRC.•Better correspondences are established between subject's PRC anatomies.•Regional and summary measures yield stronger power in discriminating aMCI.•Spatial distributions of early AD pathology may vary among anatomical variants.
