Abstract
•We propose a novel thresholding method and color transformation methods.•The G, H and S three single-channel images are considered, which improves the adaptability of the algorithm to various backgrounds.•In order to deal with the situation of connected cells, an improved concave-convex iterative repair algorithm is proposed.
