Selectively Capturing Monocyte from Whole Blood on Microfluidic Biochip for Sepsis Diagnosis
Abstract
Sepsis, a clinical syndrome caused by infection, has been frequently observed in the patients who are young, old, and with a weakened immune system. The low expression of HLA-DR (Human leukocyte antigen) on the surface of leukocyte is believed to reliably examine the sepsis patient's immune system. We report a microfluidic biochip to quickly separate leukocytes from minute whole blood sample by using deterministic lateral displacement and deposit leukocytes into antibody-coating micro-holes matrix. The monocytes are selectively captured by antibody while other leukocytes are dropped by flipping the device. Currently, our biochip can capture surface-modified THP-1 which are utilized to mimic monocytes and can selectively capture THP-1 from mixed sample of THP-1 and beads.
