Rheumatoid Arthritis Analysis by Nash Equilibrium Game Analysis
Abstract
To effectively advance the evaluation of the pharmacokinetics and pharmacodynamics characteristics in treating rheumatoid arthritis, a profile analysis method was posed to analyze the pharmacokinetics and pharmacodynamics of rheumatoid arthritis with tripterygium by Nash equilibrium game analysis. The model fits the blood concentration of hydroxytriptolide and with robust parameter estimation. Findings exhibited that the higher the level of hemoglobin (HBG), the higher clearance of hydroxytriptolide will achieve. Of efficacy, ACR20 is significantly related to the body exposure to hydroxytriptolide (P < 0.05). For safety, the correlation between body exposure to hydroxytriptolide and adverse reactions, and adverse reactions of reproductive system in women at childbearing stage was not significantly related. The established population pharmacokinetic model satisfactorily expresses the pharmacokinetic profile of hydroxytriptolide in patients. The exposure to hydroxytriptolide was significantly correlated with ACR20 and adverse reactions of the reproductive system.
